Abstract
Methysticin (CAS 495-85-2) is the major kavalactone naturally present in the roots of kava (Piper methysticum). Scientists primarily study kavalactones for their anxiolytic effects. [1] In addition, methysticin shows potent neuroprotective properties. It supports cognitive function and reduces oxidative stress in neural tissues. Methysticin also demonstrates strong anxiolytic activity, which explains kava’s traditional use for stress relief and relaxation. Furthermore, methysticin participates in anti-inflammatory pathways. Therefore, researchers consider it relevant for pain management and immune modulation studies.
Moreover, the major kavalactones (except desmethoxyyangonin) potentiate GABAA receptors. They inhibit the reuptake of norepinephrine and dopamine, bind to CB1 receptors, block voltage-gated sodium and calcium channels, and reversibly inhibit monoamine oxidase B. However, several kavalactones exhibit hepatotoxic properties, especially methysticin and yangonin. [2] These compounds also inhibit enzymes in the CYP450 family, which play a key role in drug metabolism.
