Valerenic Acid – CAS 3568-10-6

Abstract

Valerenic acid (CAS 3568-10-6) is a monocarboxylic acid and a bicyclic sesquiterpenoid phytochemical. It is naturally occurring in valerian (Valeriana officinalis), with the highest concentration in the root. Between this compound and its two naturally occurring derivatives (acetoxyvalerenic acid and hydroxvalerenic acid), valerenic acid occurs at a significantly higher concentration. In alternative medicine, extracts from valerian have served to treat various ailments for over 2,000 years. It was first utilized for treatment of a brain disorder in the late 16th century. Valerian has been and continues to be used as an herbal sedative which may be helpful in the treatment of insomnia. Furthermore, its history of use dates back to the time of ancient Greece and Rome.

This group of organic acids produce various pharmacological effects, including sedative, antidepressant, neurotoxic, sedative, and anxiolytic. Both of these acids possess roles in brain-derived neurotrophic factor (BDNF) and gastrointestinal (GIT) motility. Valerenic acid may also affect Alzheimer’s disease, through an increase in BDNF expression. The sesquiterpenoid compound is a subtype-selective GABAA receptor positive allosteric modulator via a binding site in the transmembrane domain at the β+α− interface.
